With the objective to minimize difficulties for beginners we are proposing the use of a conventional spreadsheet for the calculations of the main germination (or emergence) measurements, the organization of the final data for the statistical analysis and some electronic commands involved in these steps.

Jatropha curcas L. (Euphorbiaceae) is important for biofuel production and as a feed ingredient for animal. However, the presence of phorbol esters in the oil and cake renders the seeds toxic. The toxicity ofJ. curcas oil is currently assessed by testing in animals, leading to their death. The identification of toxic and nontoxic improved varieties is important for the safe use of J. curcas seeds and byproducts to avoid their environmental toxicity. Hence, the aim of this study was to propose a short-term bioassay using a plant as a model to screen the toxicity of J. curcas oil without the need to sacrifice any animals. ABSTRACT To support breeding programs of pear tree, on the selection of cultivars to subtropical area in Brazil, the objective of this research was to adjust the growth medium basic to pollen grain germination. ABSTRACT Pollen grain germination in vitro indicates viability and consequently provides information related to fruit set. It also assists in the development of hybrids. Along with a suitable species, a standard culture medium is essential for evaluating pollen viability. ABSTRACT The objective of this study was to identify the most efficient method for overcoming coat-imposed dormancy and determine the optimal germination temperature for Ornithopus pinnatus seeds. Treatments to overcome dormancy were: intact seeds; immersion in hot water at 60 ºC, followed by soaking in the same water (unheated)/24 h; immersion in hot water at 90 ºC, followed by soaking in the same water (unheated)/24 h; mechanical scarification; chemical scarification, H2SO4/5 min; and chemical scarification, H2SO4/10 min. The effects of temperature on germination and endo-beta-mannanase activity in seeds of Sesamum indicum was investigated. The minimum germination temperature (Tmin) lies between 12.8°C and 13.2°C while the maximum temperature (Tmax) is located between 45.5°C and 46°C. Germinabilities are statistically not different from estimated viability (88%) between 18.8°C and 43.2°C. The Mann-Whitney test indicated the interval 31.9°C to 35.1°C as the optimum temperature (Topt) range for germination rate. When seeds incubated at temperatures at or below the Tmin and close to or above the Tmax were transferred to 30°C, those incubated at lower temperatures achieved high germinability.
